In this experiment, students will isolate their mitochondrial DNA and use the Polymerase Chain Reaction (PCR) to amplify two separate regions of the mitochondrial genome. Results are analyzed using agarose gel electrophoresis.

For 25 students.
Time required: extraction 50 minutes, PCR set up 10 minutes, PCR 120 minutes, electrophoresis 60 minutes, staining 5 minutes to overnight.

All you need: DNA Electrophoresis, Thermal Cycler, Micropipettes: 5-50 µl, UV or Blue Light Transilluminator, White Light Box (optional), (2) Water Baths, Microcentrifuge & Microwave or Hot plate.
